چکیده

The soil bacterium Bacillus subtilis must cope with the environmental change including availability of nutrients, temperature, salinity, pH, antibiotics, toxins, bacteriophage, radiation, and other factors capable of damaging cells. Therefore, B. subtilis cells are equipped with a genetically regulated program to adapt to these changes. Recently, several instance of multicellular behavior of bacteria to maximize their survival are revealed. Spore formation is the ultimate surviving strategy for longtime, which is distinct physiological state form vegetative cells. Sporulation involves a highly ordered program of gene expression and morphological change. Intercellular signal transduction and compartment-specific gene expression brings about maturation of the spore. Mature spores remain for long periods of starvation and are resistant to several environmental insults. These properties are attributed mainly to the unique figures of spore’s outer layers, spore coat and cortex, as well as to the physical state of spore cytoplasm. In this chapter, we describe about surviving strategies, regulation of spatiotemporal gene expression during sporulation, and structure and resistance of the spores of B. subtilis. Sporulation in bacillus subtilis. Antigenic changes during spore formation.

1. Antisera, prepared against extracts of cells and spores of Bacillus subtilis, were used in immunoelectrophoretic studies of the changes occurring in cell extracts during the course of spore formation. 2. At least 15 antigens could be detected in vegetative-cell extracts by the antiserum prepared against cell extracts and at least seven could be demonstrated in spore extracts by the homologou...

Changes in Microsporum gypseum mycelial wall and spore coat glycoproteins during sporulation and spore germination.

Ethylenediamine-soluble glycoproteins were extracted from isolated Microsporum gypseum hyphal walls during sporulation and from spore coats before and after germination. This study was carried out to identify a sporulation-specific cell wall protein that possibly served as a substrate for the alkaline protease which initiated the macroconidial germination of this fungus. Analyses revealed that ...

Single, chemically defined sporulation medium for Bacillus subtilis: growth, sporulation, and extracellular protease production.

The composition and application of a single, chemically defined medium or growth and sporulation of Bacillus subtilis is described. At 37 degrees C cells grew with a doubling time of about 40 min; cultures attained near-maximal spore formation (70 to 80% by 12 h after the end of exponential growth and produced 1 X 10(9) to 2 X 10(9) heat-resistant free spores at 24 h. Dipicolinic acid productio...

Comparative analysis of Bacillus weihenstephanensis

22 The impact of Bacillus weihenstephanensis KBAB4 sporulation temperature 23 history was assessed on spore heat resistance, germination and outgrowth capacity at a 24 temperature range from 7 to 30°C. Sporulation rate and efficiency decreased at low 25 temperature, as cells sporulated at 12, 20 and 30°C with approximately 99% 26 efficiency, whereas at 7°C and 10°C, a maximum 15% of sporulation...
